Netherton syndrome

Netherton syndrome is a complex skin disorder primarily affecting the integumentary system while also involving hair, gastrointestinal function, and immune regulation. It is caused by pathogenic variants in the SPINK5 gene and is inherited in an autosomal recessive manner. The condition is characterized by early-onset skin abnormalities including ichthyosiform erythroderma and a distinctive hair shaft defect, along with atopic features. This mode of inheritance implies that the condition is more common when parents share a common ancestor, as both are more likely to carry the same rare genetic variant.
