Any geleophysic dysplasia in which the cause of the disease is a mutation in the FBN1 gene.
Features include always present findings: Short foot, Limitation of joint mobility, Short stature, and Short palm; and sometimes findings: Aortic valve stenosis, Enlarged liver (hepatomegaly), Tricuspid stenosis, and High blood pressure in lung arteries (pulmonary arterial hypertension) and others. 24 total HPO annotations.
Organ System | Phenotype Count | Example Features |
|---|---|---|
Heart and blood vessels | 5 | Aortic valve stenosis, High blood pressure in lung arteries (pulmonary arterial hypertension), Mitral stenosis |
FBN1 encodes fibrillin 1 (2,871 aa). Structural component of the 10-12 nm diameter microfibrils of the extracellular matrix, which conveys both structural and regulatory properties to load-bearing connective tissues. Highest expression in Cells Cultured fibroblasts (295.9 TPM) and Artery Coronary (63.8 TPM).
Geleophysic dysplasia 2 is associated with mutations in the FBN1 gene on chromosome 15.
The FBN1 protein participates in Tropoelastin associates with microfibrils and Elastic fibre formation pathways.
FBN1 is classified as a druggable target (Druggable Genome, Hormone Activity, Kinase, and Transcription Factor categories) with score 13.1.
No consensus clinical diagnostic criteria for geleophysic dysplasia have been published.
Geleophysic dysplasia should be suspected in individuals with the following clinical and radiographic findings.
Clinical findings
Proportionate short stature
Very short hands and feet

Geleophysic dysplasia is a progressive disorder resembling a lysosomal storage disorder, involving bones and joints, cardiac valves, and skin. To date about 100 individuals have been reported: 51 affected individuals in two large cohorts and 55 in case reports [, , , , , , , , , , , , , , , , , , , , , , , , , , , , , , , , , , ]. Table 2. Feature | % of Persons w/Feature | Comment |
|---|---|---|
Skeletal | Characteristic skeletal manifestations | 100% |
Additional orthopedic manifestations | 30%-40% | Osteochondritis, carpal tunnel syndrome |
Cardiac manifestations | 70% | Valvulopathy (progressive in half of affected persons); atrioventricular septal defect in 20% of individuals |
Pulmonary hypertension | 40% | Respiratory/ |
ENT | Respiratory manifestations | 50% |
Multilevel airway obstruction | 30% | Airway stenosis /or malacia |

The acromelic dysplasia group includes four rare disorders with striking clinical overlap: geleophysic dysplasia, Weill-Marchesani syndrome, acromicric dysplasia, and Myhre syndrome. Overlapping and distinguishing clinical features are summarized in . Hepatomegaly and early mortality are encountered only in the most severe forms of geleophysic dysplasia.
