Loeys-Dietz syndrome

Loeys-Dietz syndrome is a genetic connective tissue condition that primarily affects the blood vessels, skeleton, skin, and craniofacial structures. Loeys-Dietz syndrome is caused by pathogenic variants in one of several genes—SMAD2, TGFB2, SMAD3, TGFBR2, TGFB3, or TGFBR1—that encode proteins crucial for proper TGF-β signaling, a pathway that helps maintain connective-tissue strength and elasticity. Disruption of this pathway weakens the walls of blood vessels and alters skeletal and skin development, leading to the diverse clinical picture. Because the condition involves multiple organ systems, coordinated multidisciplinary care—often led by a cardiologist or geneticist familiar with connective-tissue disorders—helps ensure comprehensive monitoring and prompt intervention. Genetic counseling provides guidance on family planning and informs relatives about testing options.

Outcomes for individuals with Loeys-Dietz syndrome depend largely on the severity and timing of vascular events as well as access to specialized care. With vigilant monitoring, modern imaging techniques, and proactive surgical intervention, many people now live into adulthood with good quality of life, although lifelong follow-up remains necessary.
