IL-13 antibody REGN20423 is currently in a Phase 1 clinical trial for atopic dermatitis, and the other three antibodies are expected to enter clinical studies in 2027. Regeneron will also have the option to include in the collaboration Sanofi’s lunsekimig, an investigational bispecific Nanobody® VHH therapy targeting TSLP and IL-13, exercisable upon completion of its Phase 3 trials for chronic obstructive pulmonary disease. “This is a deliberate first step in igniting Sanofi’s innovation engine. Our longstanding Alliance with Regeneron has transformed the treatment landscape for millions of patients living with diseases driven by type 2 inflammation and created one of the world's leading immunology businesses,” said Belén Garijo, Chief Executive Officer of Sanofi. Published phase 1 findings support the promising efficacy and tolerability of anito-cel in relapsed/refractory multiple myeloma, explains lead author Matthew Frigault, MD. The phase 1 trial evaluated anito-cel, which uses a synthetic d-domain BCMA binder (ddBCMA), at 2 dose levels, 100 × 10⁶ CAR-positive T cells and 300 × 10⁶ CAR-positive T cells, in patients with relapsed or refractory multiple myeloma who had received 3 or more prior lines of therapy or had triple-class refractory disease; almost 70% were penta-drug refractory. The primary end points were adverse events during the treatment period and establishment of the recommended phase 2 dose. Among 75 of 117 treated patients who received at least 3 prior lines of therapy and were evaluated for minimal residual disease (MRD), ... At the International Myeloma Society Annual Meeting, matching-adjusted indirect comparisons comparing iMMagine-1 patients to those in pivotal trials of bispecific antibodies found higher odds of deep responses with anito-cel vs teclistamab (Tecvayli) and talquetamab (Talvey) as fourth-line treatment.4 Frigault viewed the lack of these rare but serious neurotoxicities at this longer follow-up as promising and also stressed the lack of immune effector cell–associated enterocolitis, another notable delayed complication of commercially available CAR T-cell therapy. “These were great results for a phase 1 trial,” senior author Michael R. He said that currently, CAR T–treated patients are monitored for months for neurotoxicities and enterocolitis; a therapy that avoids these toxicities can give patients more freedom and less anxiety about delayed adverse events. ... Degevma was approved for skeletal-related events in adults with bone metastases, giant cell tumor of bone, and the treatment of hypercalcemia of malignancy. ... In the phase 3 EXCALIBER-RRMM trial, iberdomide plus daratumumab and dexamethasone doubled MRD-negative complete response rates over standard therapy in relapsed myeloma.

It is currently being evaluated in a Phase II clinical trial for the treatment of Myelofibrosis. * RVU120: Ryvu Therapeutics RVU120 is an oral, selective inhibitor of CDK8 and CDK19, key regulators of transcriptional pathways involved in oncogenesis and inflammation. In Myelofibrosis, it modulates aberrant signaling such as STAT-driven pathways, helping to suppress malignant hematopoietic cell proliferation. The drug has demonstrated potential disease-modifying activity, including reduction of inflammatory cytokines and fibrosis-associated signaling. EloraTZP—a combination of an amylin asset and Eli Lilly’s approved obesity drug Zepbound—made a clean sweep of the primary and secondary endpoints in a mid-stage trial of adults with obesity and type 2 diabetes, setting up the launch of a Phase 3 trial before the end of the year.

The experimental weight-loss drug is a triple agonist and the first of its kind to reach a phase 3 clinical trial Next-generation weight-loss drugs just hit a new milestone: In a phase 3 clinical trial, retatrutide helped people with type 2 diabetes lose an average of up to 49.6 pounds, or more than 20 percent of body weight—a first for the field—according to the drug’s maker, Eli Lilly. Importantly, trial participants also saw significant reductions in blood sugar, as well as lower cholesterol and blood pressure—both of which are risk factors for cardiovascular disease. Drugs such as tirzepatide, sold as Mounjaro and Zepbound, target two receptors: GLP-1 and glucose-dependent insulinotropic polypeptide (GIP). Retatrutide targets three: GLP-1, GIP and glucagon. It’s the first such “triple agonist” to be put to the test in a phase 3 clinical trial—the gold standard test needed for the U.S. A separate clinical trial, also funded by Eli Lilly, found that participants with obesity who were not diabetic saw an up to 25 percent loss in body weight over the course of 80 weeks when they received the same doses of retatrutide. Retatrutide is not side-effect-free, however. Dosed participants in TRIUMPH-2 have reported diarrhea, constipation, nausea and vomiting. (Seven people died during the course of the study, the Lancet paper notes, but their deaths were determined to be unrelated to the treatment.)

A once futuristic-sounding cancer treatment that involves extracting a patient’s own immune cells, re-engineering them to recognize tumor targets, and returning them to the body is beginning to move beyond oncology. At Yale School of Medicine (YSM), physician-scientists are testing CAR-T (chimeric antigen receptor-T cell) therapy in clinical trials for autoimmune diseases including lupus, multiple sclerosis (MS), and scleroderma. Yale researchers are testing CAR-T therapy in clinical trials for autoimmune diseases including lupus, multiple sclerosis, and scleroderma. They're also working Teams from hematology-oncology, rheumatology, and neurology have come together to study CAR-T therapy for MS, scleroderma, and lupus in clinical trials. The program launched in August 2025 when the first patient with MS received CAR-T cells. Earlier this year, Yale clinicians performed the first CAR-T infusion for lupus in the state. ... Multiple sclerosis is an immune-mediated disease in which the body’s own immune cells mistakenly attack the brain and spinal cord, damaging the protective myelin that allows nerve cells to communicate. That limitation is central to the work of Yale’s Erin Longbrake, MD, PhD, associate professor of neurology and principal investigator of an early-phase clinical trial testing whether CAR-T therapy can eliminate disease-driving immune cells not only in the bloodstream, but throughout the body—including the brain.

A real-world study found that adding a GLP-1 to SGLT2 inhibitor therapy was associated with lower kidney, cardiovascular, and mortality risks in CKD. Observational design, potential immortal-time bias, residual confounding, limited dosing/adherence data, and short follow-up constrain causal inference, reinforcing the need for confirmatory randomized trials.SHOW MORE · A real-world study found that adding a GLP-1 to SGLT2 inhibitor therapy was associated with lower kidney, cardiovascular, and mortality risks in CKD. ... Adding a glucagon-like peptide-1 receptor agonist (GLP-1 RA) to ongoing SGLT2 inhibitor therapy was associated with lower risks of kidney disease progression, The FDA has approved belzutifan plus lenvatinib for advanced ccRCC after PD-1/PD-L1 therapy, expanding treatments for those with disease progression. "The addition of a GLP-1 RA to ongoing SGLT2 [inhibitor] therapy is associated with a substantial reduction in the risk of kidney disease progression and cardiovascular events in a real-world setting,” they wrote. "…While confirmatory randomized trials are awaited, these data support the early adoption of combination therapy to address the high residual risk in patients with CKD.” The primary outcome was major adverse kidney events (MAKE), a composite of all-cause mortality, progression to end-stage kidney disease (ESKD), dialysis, stage 5 CKD, or kidney transplant. Over a median follow-up of 12 months, combination therapy was associated with a lower risk of MAKE compared with SGLT2 inhibitor monotherapy (HR, 0.70; 95% CI, 0.65-0.74; P < .001).
