Any megalencephaly-polymicrogyria-postaxial polydactyly-hydrocephalus syndrome in which the cause of the disease is a mutation in the PIK3R2 gene.
Features include always present findings: Hydrocephalus, Enlarged brain ventricles (ventriculomegaly), and Polymicrogyria; and sometimes findings: Overgrowth and Postaxial polydactyly. 27 total HPO annotations.

MPPH syndrome is a developmental brain disorder characterized by megalencephaly (brain overgrowth) with the cortical malformation bilateral perisylvian polymicrogyria. To date, fewer than than 100 individuals with features of MPPH syndrome have been reported with either a clinical diagnosis (presence of the 2 core clinical and imaging findings: megalencephaly and polymicrogyria) , and/or a molecularly confirmed diagnosis [, , , , , , , , , , , , , , , , , , , ]. Megalencephaly-polymicrogyria-polydactyly-hydrocephalus syndrome 1 is associated with mutations in the PIK3R2 gene on chromosome 19.
In general, no differences in phenotype have been observed between individuals with a molecularly confirmed diagnosis and those with only a clinical diagnosis. The exceptions are several individuals with a molecularly confirmed diagnosis of MPPH syndrome who had BPP but lacked the core clinical feature of megalencephaly .
