MPPH syndrome is a developmental brain disorder characterized by megalencephaly (brain overgrowth) with the cortical malformation bilateral perisylvian polymicrogyria. To date, fewer than than 100 individuals with features of MPPH syndrome have been reported with either a clinical diagnosis (presence of the 2 core clinical and imaging findings: megalencephaly and polymicrogyria) , and/or a molecularly confirmed diagnosis [, , , , , , , , , , , , , , , , , , , ]. CCND2 encodes cyclin D2 (289 aa). Regulatory component of the cyclin D2-CDK4 (DC) complex that phosphorylates and inhibits members of the retinoblastoma (RB) protein family including RB1 and regulates the cell-cycle during G(1)/S transition. Highest expression in Cells EBV-transformed lymphocytes (134.5 TPM) and Colon Sigmoid (76.3 TPM).
Megalencephaly-polymicrogyria-polydactyly-hydrocephalus syndrome 3 is caused by mutations in the CCND2 gene on chromosome 12.
The CCND2 protein participates in p-T172-CDK4:CCND2:CDK4 inhibitors (CCND2), p-T177-CDK6:CCND2:CDK6 inhibitors (CCND2), and Association of INK4 family proteins with CDK4/6 pathways.
CCND2 is classified as a druggable target (Clinically Actionable, Druggable Genome, Kinase, and Transcription Factor categories) with score 0.9.
In general, no differences in phenotype have been observed between individuals with a molecularly confirmed diagnosis and those with only a clinical diagnosis. The exceptions are several individuals with a molecularly confirmed diagnosis of MPPH syndrome who had BPP but lacked the core clinical feature of megalencephaly .
